One of the pieces of art I commissioned was removed in 2016 at the artist's request since they were "no longer drawing adult art and would no longer be using e621." That artist has since been removed from the avoid posting list and has art, including adult art, being uploaded to the site. Would it be acceptable for me to re-upload the piece that was previously deleted since the artist has clearly changed their mind regarding their artwork?

Do not reupload anything that was previously deleted. The only way to restore them is if the artist asks for those posts to be reinstated.

You should get the post number and ask the artist if they would be okay with it being reinstated.

I don't really care enough to bother the artist over a piece that's like 12 years old at this point. I believe all their old art was mass-deleted when they asked to be placed on the avoid posting list, so I assumed it would be okay to upload it again after they were removed from the list. If it requires explicit permission, it's not really worth the trouble. People can just see it on my FA page lol.

Don't try to reupload it, you wouldn't be able to do so anyways since the artwork's unique identifier (MD5 hash) is saved and the site would prevent you from posting something that already exists (even when it is in the deleted state).
If you alter the original file in some way and get it posted again, it may get flagged as previously-deleted content by a janitor and you would get smacked for Posting Abuse.

We are very particular with the takedown instructions.
If the artist asks for all art to be deleted and to be placed on the DNP list, we would oblige.
If the artist then asks for them to be removed from the DNP, but fail to mention restoring the artworks, we would only lift the DNP and leave the deleted artwork as is.

If you poked the artist respectfully about it, then they may approach an admin themselves and ask for their previous works to be restored.
Alternatively, they may choose not to restore it for a different reason now (e.g., don't like old art) and choose to leave things as is.
